ThioGlo® 

Superior Fluorescent Thiol Reagents

ThioGlo® maleimide reagents are superior to existing thiol reagents for detection, determination and derivatization of thiol groups using fluorescence techniques. They have little or no fluorescence prior to reaction with thiols and high quantum yields after reaction with active -SH groups in proteins, enzymes, and simple peptides resulting in extraordinary thiol sensitivity. Their reactions with thiols in aqueous buffered solutions (pH 7) are extremely fast at room temperature, usually complete in less than a minute. These reaction rates are 1000 times faster than those of bimanes under the same conditions. ThioGlo® 1 and ThioGlo® 5 are 2 to 3 times more resistant to hydrolysis of the maleimide ring in aqueous buffer than 7-diethylamino-3-(4'-maleimidyl phenyl)-4-methyl coumarin (CPM) or fluorescein maleimide (FM). Even more important is the fact that their thiol inactive hydrolysis products are practically nonfluorescent, and thus do not interfere with thiol adduct fluorescence. This allows quantitation without separation from either the reagent or the hydrolysis product. ThioGlo® 3 is most suitable for intracellular determination of GSH and other thiols in viable cells because of its lower cytotoxicity. All ThioGlo® reagents have large Stokes shifts, and rather broad excitation and emission bands (allowing use of broad band filters) and are far more resistant to photobleaching than fluorescein.
